FDA Issues Draft Guidance on BLA Submissions for Individualized Genome Editing Products
FDA's CBER releases draft guidance on single-BLA submissions for individualized genome editing products, with comments due 27 November 2026.

A regulatory framework shift for advanced therapy manufacturers arrived on 27 October 2026, when FDA's Center for Biologics Evaluation and Research (CBER) published draft guidance addressing how individualized genome editing (GE) products, including those spanning multiple product variants, can be submitted under a single Biologics License Application (BLA) pursuant to section 351(a) of the PHS Act. The comment window closes 27 November 2026, leaving CMC, QA, and regulatory affairs teams a narrow four-week window to shape the final version.
The guidance targets sponsors developing individualized GE products and covers four operationally significant areas: specifying the permissible extent of GE component variation within a BLA; adding new product variants postapproval; in vitro safety and potency evaluation of those variants; and manufacturing process validation requirements. For QA directors, the postapproval variant management provisions carry the most immediate weight, the framework will determine how tightly CMC strategies must be scoped at licensure and how much flexibility remains for variant additions without triggering a full supplemental BLA.
The process validation language is particularly consequential for plant heads operating individualized therapy platforms. Unlike conventional biologics, individualized GE products present a moving target: each patient-specific variant may introduce manufacturing variables that existing validation protocols under 21 CFR Part 211 and ICH Q10 quality system principles were not designed to accommodate at scale. The guidance signals that FDA expects sponsors to define the boundaries of acceptable variation upfront, with a documented rationale that can withstand postapproval inspection scrutiny.
Labeling, indication statements, and drug naming and identification are also addressed, areas where regulatory leads will need to coordinate closely with clinical and commercial teams to ensure consistency across variant-specific labeling without creating a compliance gap at the product level. The docket number for written submissions is FDA-2026-D-11783, and CBER's Office of Therapeutic Products has published a recorded webinar featuring Andrew Byrnes, PhD and Jacob Bitterman, PhD to provide a high-level orientation to the document.
The degree to which sponsors engage during this comment period will directly influence how prescriptive the final guidance becomes on variant boundary definitions and postapproval change management pathways.
