Lexicon Secures Third $10M Milestone from Novo Nordisk for Phase 1 Dosing

A patient dosing milestone in Novo Nordisk's Phase 1 program for LX9851 has triggered a third $10 million payment to Lexicon Pharmaceuticals in 2026, bringing cumulative milestone receipts under the collaboration to $75 million against a potential $1 billion ceiling. For manufacturing and regulatory teams tracking the oral obesity pipeline, the pace of clinical progression signals that non-incretin small molecule candidates are moving faster than many anticipated.

LX9851 is a potent, selective oral inhibitor of Acyl CoA Synthetase 5 (ACSL5), a target implicated in fat accumulation and energy balance. Novo Nordisk secured an exclusive worldwide license to develop, manufacture, and commercialize the compound across all indications under an agreement signed in 2025. The Phase 1 program, initiated in March 2026, is evaluating safety, tolerability, pharmacokinetics, and pharmacodynamics of single and multiple ascending doses versus placebo in overweight and obese adults, with completion expected in Q1 2027.

The clinical profile carries upstream manufacturing relevance. Unlike GLP-1 receptor agonists requiring complex biologics infrastructure, an oral small molecule like LX9851 sits within conventional solid-dose manufacturing frameworks governed by 21 CFR Part 211 and ICH Q10 quality system requirements. Should the compound advance through later-stage development, process validation and scale-up planning for oral dosage forms will need to begin well ahead of pivotal trial supply commitments.

Preclinical data presented at Obesity Week 2024 showed LX9851 combined with semaglutide produced greater reductions in weight, food intake, and fat mass than semaglutide alone. Separately, the compound demonstrated mitigation of weight regain and positive effects on liver steatosis following semaglutide discontinuation in animal models. These combination and sequencing signals may shape how Novo Nordisk structures future clinical supply chains if LX9851 progresses alongside existing GLP-1 assets.

Under the collaboration terms, Lexicon retains eligibility for tiered royalties on net sales in addition to the remaining milestone payments.
