Achieve Life Sciences Resubmits Cytisinicline NDA After cGMP-Driven CRL Forces CMO Transition
Achieve Life Sciences targets Q4 2026 NDA resubmission after a cGMP-driven CRL forced a finished drug product CMO transition to Adare Pharma Solutions.


A Complete Response Letter tied entirely to manufacturing deficiencies, not clinical data, is reshaping Achieve Life Sciences' path to U.S. approval for cytisinicline, and the company's response offers a direct case study in managing CMO transitions mid-approval cycle. The CRL, issued June 20, 2026, cited outstanding cGMP observations from an inspection of the prior third-party finished drug product facility, which received an Official Action Indicated classification, alongside incomplete final product labeling. FDA identified no deficiencies in clinical efficacy or safety.
For QA directors and regulatory leads, the sequence matters: the company disclosed in April 2026 that a CRL was anticipated, signaling that the OAI classification at the original CMO was already an active risk before the letter arrived. The manufacturing transition to Adare Pharma Solutions, a U.S.-based contract manufacturer, was initiated in parallel. Achieve has since completed analytical method technology transfer, manufactured its first engineering batch at Adare's facility, and fully qualified testing procedures there, steps that map directly to 21 CFR Part 211 site change requirements and ICH Q10 quality system expectations for post-approval manufacturing changes.
The NDA resubmission, planned for Q4 2026, will name Adare as the commercial finished drug product manufacturer. FDA approval is anticipated in the first half of 2027. To support the resubmission and commercial readiness, Achieve appointed Ronald Dadino as Senior Vice President, CMC and Supply Chain, and Gloria Cosgrove as Senior Vice President, Global Quality, both carrying U.S.-based pharmaceutical manufacturing credentials. The commercial team was similarly expanded, with hires drawn from the Ohtuvayre launch at Verona Pharma.
Financing is structured around the regulatory timeline: a private placement closed at up to $354 million, with $180 million upfront and up to $174 million from warrants exercisable prior to and up to 20 trading days following FDA approval. Six new directors joined the board, adding healthcare investment, biopharmaceutical finance, and clinical-stage operational depth.
On the clinical side, 52-week safety data from the ORCA-OL study were presented at the American Thoracic Society 2026 Annual Meeting, completing the anticipated data package for cytisinicline's long-term safety profile in nicotine dependence.
The Q4 2026 resubmission date, and whether Adare's facility clears any subsequent FDA inspection without further observations, will be the measurable checkpoint that determines whether the first-half 2027 approval window holds.
